Phenotypical and functional alterations in the mucosal immune system of CD45 exon 9 KO mice.

چکیده
The protein tyrosine phosphatase CD45 has been shown to be an important regulator of antigen-receptor signaling in both T and B lymphocytes. Lymphocyte populations within the gut mucosa are phenotypically and ontogenetically different from those generally found in secondary lymphoid tissue. In T lymphopenic mice, extrathymic T cell development takes place within the gut. Here we report the characterization of T and B cell populations in the distinct compartments of the gut mucosa and gut-associated lymphoid tissue of CD45-null mice. These data suggest that CD45 is required for the development of the specialized T cell populations within the gut environment as has been previously shown for thymic development. We demonstrate that within the large intestine intraepithelial compartment alphabeta-TCR+ CD4+ T cells are selectively retained by CD45KO mice. T cells and NK cells within the intraepithelium and the gut mucosa associated lymphoid tissue of CD45KO mice frequently possess an activated phenotype, differentiated to produce typical TH1 and TH2 cytokines. These data demonstrate that local environmental differences within the gut can, at least in part, overcome the requirement for CD45 during activation of T cells.

Changes in vascular reactivity of the coronary artery and thoracic aorta in the delta sarcoglycan null mutant mice
Introduction: Mutations in the delta sarcoglycan gene (d-SG) cause limb-girdle muscular dystrophy type 2F with structural and functional alterations in cardiac, smooth and skeletal muscle. The objective of the present study was to improve information about changes in vascular reactivity of the thoracic aorta and the coronary artery in the perfused heart of the d-SG-null mutant mouse model. ...
متن کاملO24: Functional Role of the K2P Potassium Channel TASK-3 in Glioma
TASK-3, a two-pore-domain (K2P) potassium channel, has been implicated as important regulator for the effector function and proliferation of T-cells. Interestingly, TASK-3 has also a functional impact on tumor cells. Therefore, we sought to investigate whether TASK3 modulation might have a therapeutic potential for malignant gliomas by a variety of phenotypical and functional in vitro assays mi...
متن کاملA high-frequency polymorphism in exon 6 of the CD45 tyrosine phosphatase gene (PTPRC) resulting in altered isoform expression.
CD45 (leukocyte common) antigen is a hemopoietic cell-specific tyrosine phosphatase essential for antigen receptor-mediated signaling in lymphocytes. The molecule undergoes complex alternative splicing in the extracellular domain, and different patterns of CD45 splicing are associated with distinct functions. Lack of CD45 leads to severe combined immunodeficiency, and alterations of CD45 splici...
متن کاملPolymorphisms within Exon 9, But Not Intron 8, of the Vitamin D Receptor Gene Are Associated with Asthma
Objective(s) Deregulation of the immune system through allied factors and cytokine responses are thought to be important contributors to the pathogenesis of asthma. Vitamin D3 and its nuclear receptor appear to be factors that maybe involved in regulating immune responses during the progression of asthma. The aim of this study was to investigate the association between polymorphisms in intron ...
متن کاملCombinations of CD45 isoforms are crucial for immune function and disease.
Expression of the CD45 Ag in hemopoietic cells is essential for normal development and function of lymphocytes, and both mice and humans lacking expression exhibit SCID. Human genetic variants of CD45, the exon 4 C77G and exon 6 A138G alleles, which alter the pattern of CD45 isoform expression, are associated with autoimmune and infectious diseases. We constructed transgenic mice expressing eit...
